Scientists have discovered why antidepressants do not help one third of people suffering from severe forms of depression.
We are talking about the selective inhibitors of serotonin reuptake (SSRI), which usually ease the symptoms of mood disorders, Molecular Psychiatry says.
Researchers from the Salk Institute examined the range of responses to SSRI therapy from 803 patients with severe depressive disorder.
Symptoms of depression
From this group they selected three patients in whom the observed weakening of the symptoms of depression after taking the antidepressant, and three patients who did not respond to SSRI therapy.
The researchers isolated skin cells of these patients and three healthy subjects from the control group. Physicians used methods of reprogramming stem cells to transform skin cells into pluripotent stem cells and then to neurons.
Scientists have studied how the neurons derived from each person reacted to the increased level of serotonin. When was production of the hormone, some neurons showed higher activity in comparison with the neurons of healthy people.
Causes of depression
Further experiments pointed to two specific receptor of serotonin (out of seven known in the human brain) 5-HT2A and 5-HT7. When these receptors were blocked chemical compound, the neurons ceased to be hyperactive in the presence of serotonin. This suggests that drugs aimed at the above receptors may be an effective alternative to current antidepressants in some patients, but more research is needed.
